Effect of urban and industrial expansion on the genetic biodiversity of two desert animal species in Kuwait as determined by RAPD-PCR

 ABSTRACT

The study applied molecular genetics techniques to determine the effect of urbanization and industrial expansion on the biodiversity of two desert animals Jaculus jaculus and Uromastyx microlepus.  Animals were randomly captured from three areas in Kuwait: Subiya, Jlaiya and Chebd.  Manmade and natural barriers spatially separate the areas.  

 RAPD-PCR technique was used for the rapid identification of genetic markers. A number of monomorphic and polymorphic bands were obtained using three decamer primers. The monomorphic markers were species-specific. From the analysis of monomorphic and polymorphic markers and their dissimilarity indices (DI), the three different populations of both species, J. jaculus and U. microlepus, were found to be genetically different by varying degrees. This was attributed to the geographical, man-made barriers and the life style of each species. The populations of both J. jaculus and U. microlepus in Jlaiya had a DI of 0.4 and 0.18 respectively indicating low dissimilarity.  While populations of both J. jaculus and U. microlepus of the more open area, Chabd, had a higher DI of 0.66 and 0.36 respectively. Furthermore, J. jaculus populations were found to show more biodiversity than the U. microlepus populations in a given area.
